چکیده
5. Anaerobic incubation caused an inhibition of synthesis only in the presence of oxaloacetate; no inhibition was observed anaerobically in the presence of a-oxoglutarate plus malonate. 6. Hg2+ and arsenate almost completely abolished fatty acid synthesis; cyanide and 2:4-dinitrophenol caused a moderate (30-70%) inhibition, while azide and fluoride were without effect. 7. The implication of the results is discussed.

Biosynthesis of Fatty Acids I. STUDIES WITH ENZYMES OBTAINED FROM LIVER
The requirement for bicarbonate in the incubation medium for the conversion of octanoic acid to stearic acid by slices of liver tissue was reported some 10 years ago (1). Furthermore, the inclusion of bicarbonate in the homogenizing medium permitted the preparation of a consistently active enzyme system from pigeon liver which catalyzed the conversion of acetate or acetyl coenzyme A to long cha...
متن کاملPurification and some properties of a medium-chain acyl-thioester hydrolase from lactating-rabbit mammary gland which terminates chain elongation in fatty acid synthesis.
1. An acyl-thioester hydrolase was isolated from the cytosol of lactating-rabbit mammary gland. The purified enzyme terminates fatty acid synthesis at medium-chain (C8:0-C12:0) acids when it is incubated with fatty acid synthetase and rate-limiting concentrations of malonyl-CoA. These acids are characteristic products of the lactating gland. 2. The mol.wt. of the enzyme is 29000+/-500 (mean+/-S...
متن کاملThe Conversion of Long Chain Saturated Fatty Acids to Their @Unsaturated, ,&y-Unsaturated, and @-Hydroxy Derivatives by Enzymes from the Cellular Slime Mold, Dictyostelium discoideum
A major problem in the study of fatty acid metabolism at the subcellular level has been the demonstration of the individual steps of both oxidation and synthesis (1, 2). Oxidation of long and short chain fatty acids by intact mitochondria is considered to proceed directly to acetate, without the accumulation of intermediates (3). Synthesis of fatty acids from acetyl coenzyme A and malonyl-CoA b...
متن کاملFat metabolism in higher plants. XIV. Factors affecting the synthesis of oleic acid by particulate preparations from avocado mesocarp.
Numerous investigations have led to the point at which refined enzyme systems may be used to study the biosynthesis of saturated fatty acids (2, 3). The biosynthesis of unsaturated fatty acids has not been so thoroughly elucidated. Formation of unsaturated fatty acids has been observed experimentally in viva (4-9) and in vitro (10, 11) but several of these studies showed that incorporated radio...
متن کاملIdentification of an acyl donor in steryl ester biosynthesis by enzyme preparations from spinach leaves.
A pathway for steryl ester biosynthesis in acetone powder preparations from spinach (Spinacia oleracea L.) leaves has been elucidated; free sterol and 1,2-diglyceride were the substrates. Although animals synthesize cholesteryl esters by three distinct biosynthetic pathways, none of these pathways utilizes 1,2-diglyceride as an acyl donor. Phosphatidylcholine, phosphatidic acid, triglyceride, 1...
